Abstract

Overall genome relatedness index (OGRI) methods calculate the sequence-level similarity of genomes as “percentage identity,” and OGRI scores are often interpreted to represent the true proportion of identical, evolutionarily-equivalent sites in each genome. But is that actually the case? Here we present findings from the first objective benchmarking of OGRI methods against a synthetic dataset emulating point mutation and structural variation. We find that the methods tested have different ranges of accuracy and applicability, but are generally robust to genome structural variation, and we make recommendations for use and interpretation of OGRI method outputs.
